Environmentally-Friendly Workflow Based on Supercritical Fluid Chromatography and Tandem Mass Spectrometry Molecular Networking For the Discovery of Potent Anti-Viral Leads From Plants

چکیده
A supercritical fluid chromatography-based targeted purification workflow using tandem mass spectrometry and molecular networking was developed to analyze, annotate and isolate secondary metabolites from complex mixture. This approach was applied for targeted isolation of new antiviral diterpene esters from Euphorbia semiperfoliata whole plant extract. The analysis of bioactive fractions revealed that unknown diterpene esters, including jatrophane esters and phorboids esters, were present in the samples. The purification procedure using semi-preparative-supercritical fluid chromatography led to the isolation and identification of two jatrophane esters (13 and 14) and four 4-deoxyphorbol esters (15-18). Compound 16 was found to display antiviral activity against chikungunya virus (EC50 = 0.45 μM), while compound 15 was found to be a potent and selective inhibitor of HIV-1 replication in a recombinant virus assay (EC50 = 13 nM). This study showed that supercritical fluid chromatography-based workflow and molecular networking can facilitate and accelerate the discovery of bioactive small molecules by targeted molecules of interest, while minimizing the use of toxic solvents. not peer-reviewed) is the author/funder. An Environmental Friendly Process for Extraction of Active Constituents from Herbal Plants
Conventional methods for extraction of the active constituents from herbal plants, in addition to their high processing costs, are harmful to the environment owing to their time and energy consumption, uncontrollable parameters, use of toxic and dangerous solvents, loss of the extract, thermal instability, the contamination of the product with organic solvents and the loss of water-soluble comp...
متن کاملIdentification and quantification of cannabinoids in Cannabis sativa L. plants by high performance liquid chromatography-mass spectrometry.
High performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS) has been successfully applied to cannabis plant extracts in order to identify cannabinoid compounds after their quantitative isolation by means of supercritical fluid extraction (SFE). MS conditions were optimized by means of a central composite design (CCD) approach, and the analysis method was fully validated. Six ma...

متن کاملSimultaneous Analysis of Metabolite Residues from Degradation of Dithiocarbamates in Cereals Using Liquid Chromatography Coupled with Tandem Mass Spectrometry
Background and Objectives: Dithiocarbamates are groups of fungicides used to control pests of agricultural products. Analysis of these pesticides with liquid and gas chromatography instruments is associated with various problems due to the presence of metal groups in the structure of these compounds. Therefore, one of the ways for analysis the pesticides is to check the level of decomposition...
متن کاملComparative Pharmacokinetics of Berberine, Palmatine and Jatrorrhizine in Rat Plasma after Oral Administration of Rhizoma coptidis and Zuojinwan Using Liquid Chromatography-Tandem Mass Spectrometry
A selective and sensitive liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed for the determination of berberine, palmatine and jatrorrhizine in rat plasma. Target compounds, together with the internal standard (metronidazole), were extracted from rat plasma samples by protein precipitation with acetonitrile-methanol (1:2, v/v). Chromatography was carried out using a ...
